This candidate has knowledge of confocal microscopy, optical design, (Matlab) programming, signal processing, FPGA programming. The candidate has a strong affection for biological research and has the ability to work in a team and also independently.

Project description: Ultra-Sensitive Confocal Microscopy
Description:
Confocal microscopy is a technology to make 3D images of fluorescently labelled biological objects. Since the first development of the confocal microscope in the early 80s the detection efficiency has been improved substantially by using more sensitive detectors, better optical design and improvements of the acquisition electronics. It is the aim of this (STW funded) project to further improve the sensitivity of the microscope. To do that we will exploit the novel technology ‘Controlled Light Exposure Microscopy' (CLEM), invented and developed by Prof. E. Manders and Dr R. Hoebe. CLEM has been designed for reduction of photodamage in fluorescence live-cell microscopy. In this project CLEM will be exploited to improve detection sensitivity. It is our aim to improve sensitivity by a factor of 50 to 100.
The PhD student will design, build and test new electronics for the image acquisition using new CLEM strategies. He/she will focus on electronic implementation and computer simulations. An important part of the PhD study will concern the characterization of the new method with respect to noise, resolution and photon statistics. This project will be supervised by Dr Ron Hoebe at the AMC location of LCAM.
Conditions of employment
The full-time appointment will be on a temporary basis for a maximum period of four years (18 months plus a further 30 months after a positive evaluation) and should lead to a dissertation (PhD thesis). An educational plan will be drafted that includes attendance of courses and (international) meetings. The PhD student is also expected to assist in teaching of undergraduates. Based on a full-time appointment (38 hours per week) the gross monthly salary will range from €2042 in the first year and €2612 in the final year, according to the Dutch salary scales for PhD students. The successful candidate will be appointed by the FNWI and will be hosted in the AMC and FNWI. The collective employment agreement (CAO) of Dutch universities is applicable.
