Description:
We have recently developed a novel technique for subtle gene modification in mouse embryonic stem cells that makes use of short single-stranded DNA oligonucleotides. We now wish to develop this technology for use in induced pluripotent stem cells (iPSC) of human origin. The graduate student will participate in the derivation and culturing of iPSC and the development of a protocol for oligonucleotide-directed gene modification in iPSC. Subsequently, the graduate student will use the technology to study the consequences of subtle variants of DNA repair genes found in the human population for development of cancer.
