Spinocerebellar ataxia (SCA) is a progressive neurodegenerative disorder and is genetically very heterogeneous.
Description:
Spinocerebellar ataxia (SCA) is a progressive neurodegenerative disorder and is genetically very heterogeneous. Despite the fact that many disease genes are known, some 30% of the ataxia patients can still not be genetically diagnosed. Here we propose to use whole exome sequencing. a next generation sequencing technology, to identify novel SCA genes in families in whom no genetic diagnosis could be made so far. We have recently developed an ataxia gene-gene interaction network. This will allow us to immediately select for those variations in genes residing in this network and identified by exome sequencing. We can thus use them to predict which genes participate in the common ataxia pathways and to highlight some of the emerging pathways leading to cerebellar neurodegeneration. In addition, potentially disease-causing variations will be functionally screened in cell- and Drosophila models. The postdoc will be responsible for the daily coordination of the project. In collaboration with the genome facility, the postdoc will prepare the DNA libraries, analyze the exome sequence data, and perform the functional assays in order to identify disease-causing variations underlying SCA.
